Yes. assuming of course that you meant sending both your vocals & midi song's audio playback out thru your PSR3000's L/R audio outputs into your computer's soundcard audio input(s).

Because this is a 'multi-track' audio recording program, for better recording control, I suggest recording your midi backing tracks and vocals in separate takes (and on separate tracks), to allow the ability for post mixing.
In addition to audio recording, PowerTrack Audio Pro 10 also supports midi editing, so if you needed to edit the midi backing track, instead of simply playing back the midi backing track thru the PSR3000's midi player, you could instead load the midi file into PowerTracks Pro Audio, and simply utilize the PSR3000 as a sound module only, affording far more midi file editing ease. In addition PowerTracks provides more flexibility and power at mixdown CD or MP3. $49, what a deal !
