I presume when you say tune your keyboard/vocals, you mean EQ. Everyone's set up, adjustments will differ based on what equipment they use.

I use the following:

Tyros
Behringer PMH 200 power mixer
Digitech Vocalist VR
Shure Beta 57A or Shure SM58 mic
2-15" EV Force 1 2-way cabinets

After getting everything set up I typically walk out front and verify my vocal. I'm looking for a pleasing sharp sound. I like to boost the lows and the highs a bit to add definition. I test the mic the same way I'll use it when I actually start playing. I sing right on the mic itself...usually touching my lower lip as a base point for output. Once I'm happy with the basic sound...I'll adjust the reverb/delay settings to a pleasant level and I'm done with it.

Next is the keyboard...Since I love how the Tyros sounds in the first place...I rarely do any major adjusting on the power amp. Depending on the room, I may cut my low end slightly...

I'll play a few bars of differing styles soft ballads, upbeat pop.country stuff...to try and get a bead on how they'll sound in performance. This usually takes less than 3-4 minutes to get the Tyros sounding just the way I want it to.

Next, I'll listen to my vocals with the harmonizer while playing softly to make sure the Digitech is set up right. Again...this usually takes less than a minute to tweak it the way I want. The main thing I'm on the outlook for is if my output control is too high on the harmonizer which triggers a red signal.

After that...I'm ready to roll...

It can take as little as a few minutes to have everythign sounding good up to 15-20 if the room is oddly shaped or I'm in a corner or something.
